dc.description.abstractThe long-term storage of spermatozoa requires presence of appropriate conditions among which is a buffer and an antibiotic to regulate ph. A number of antibiotics and buffers have been evaluated and found to be effective. However, with indiscriminate usage of antibiotics that has resulted into super resistant bacteria, there is need to constantly evaluate the efficacy of some of these antibiotics especially those which have been commonly used in humans and animals among which is ciprofloxacin. Moreover, the efficacy of ciprofloxacin varies from one ph to another thus the need to evaluate its performance in different pH ranges in such circumstances. The objective of this project was to investigate the effect of different concentrations of ciprofloxacin and sodium citrate buffer in egg yolk extender on microscopic parameters. of bovine sperm. The testes from which sperm samples were collected were obtained from Kalerwe abattoir slaughter house along Gayaza road, in Kawempe division Kampala district. Sperm samples were collected by epididymal retrograde flushing and a total of 120 observations were analyzed. The epididymal sperm samples were divided into two experiments, with experiment one testing for an increase in ciprofloxacin level against constant sodium citrate buffer concentration and the other testing for both increments in ciprofloxacin and sodium citrate buffer concentrations frozen at 4oc for three days before investigation to evaluate whether the varied contents of the egg yolk citrate had any effects on the bovine sperm parameters during storage. The analysis of the results using Microsoft excel spread sheet programs and repeated measures ANOVA using STATA software version 12.0 using a 95% confidence interval indicated that in experiment one, there was adverse and evident effect on the bovine sperm progressive motility and vitality as it presented significantly (p< 0.05) decreasing bovine parameters while in experiment two, there was a slight decrease in the bovine sperm parameters with no statistical significance (p> 0.05). It can be concluded from this study that pH evidently affects sperm parameters. However, it is recommended that more research should be done on the effect of other different antibiotics on the bull epididymal sperms under various storage treatments.en_US
